PPARδ is a nuclear receptor involved in the regulation of lipid metabolism and substrate utilisation. Agonists of this receptor alter the transcription of genes governing how tissues metabolise fatty acids and glucose, which is why the pathway attracted pharmaceutical interest for dyslipidaemia and metabolic syndrome.

GW-501516 was developed as a selective PPARδ agonist by GlaxoSmithKline in collaboration with Ligand Pharmaceuticals, and progressed through early clinical evaluation for lipid disorders. Preclinical work also examined its effects on endurance and substrate utilisation in animal models, which is the origin of the "Endurobol" nickname and much of its subsequent notoriety.

Development was discontinued in 2007 after long-duration rodent studies at high doses reported carcinogenic findings.
